Characteristics of microRNAs in Skeletal Muscle of Intrauterine Growth-Restricted Pigs

چکیده
microRNAs are a class of small RNAs that have been extensively studied, which involved in many biological processes and disease occurrence. The incidence intrauterine growth restriction is higher mammals, especially multiparous mammals. In this study, we found the weight longissimus dorsi growth-restricted pigs was significantly lower than normal pigs. Then, pig were used to characterize miRNA expression profiles by RNA sequencing. A total 333 miRNAs identified, 26 differentially expressed. Functional enrichment analysis showed these expressed regulate their target genes (such as PIK3R1, CCND2, AKT3, MAP3K7), play an important role proliferation differentiation skeletal muscle through signaling pathways such PI3K-Akt, MAPK, FoxO pathways. Furthermore, miRNA-451 upregulated IUGR muscle. Overexpression miR-451 C2C12 cells promoted Mb, Myod, Myog, Myh1, Myh7, suggesting may be regulation myoblastic cells. Our results reveal regulating myogenic with restriction.

To determine the molecular mechanism(s) linking fetal adaptations in intrauterine growth restriction (IUGR) to adult maladaptations of type 2 diabetes mellitus, we investigated the effect of prenatal seminutrient restriction, modified by early postnatal ad libitum access to nutrients (CM/SP) or seminutrient restriction (SM/SP), vs. early postnatal seminutrient restriction alone (SM/CP) or contr...
متن کاملIntrauterine Growth Retardation Increases the Susceptibility of Pigs to High-Fat Diet-Induced Mitochondrial Dysfunction in Skeletal Muscle
It has been recognized that there is a relationship between prenatal growth restriction and the development of metabolic-related diseases in later life, a process involved in mitochondrial dysfunction. In addition, intrauterine growth retardation (IUGR) increases the susceptibility of offspring to high-fat (HF) diet-induced metabolic syndrome. Recent findings suggested that HF feeding decreased...

Background: The aim of this study was to investigate the response of ERK1/2 protein and muscular morphological adaptations to a period of resistance training with local blood flow restriction. Materials and methods: Twenty healthy male Wistar rats without clinically evident disease (5 weeks old, 120±7 g weight) were divided into four equal groups: control, control with limited blood flow...
متن کاملRole of microRNAs in skeletal muscle hypertrophy
Skeletal muscle comprises approximately 40% of body weight, and is important for locomotion, as well as for metabolic homeostasis. Adult skeletal muscle mass is maintained by a fine balance between muscle protein synthesis and degradation. In response to cytokines, nutrients, and mechanical stimuli, skeletal muscle mass is increased (hypertrophy), whereas skeletal muscle mass is decreased (atro...
متن کاملMicroRNAs in the regeneration of skeletal muscle.
MicroRNAs (miRNAs) have emerged as critical regulators of numerous biological processes by modulating gene expression at the post-transcriptional level. The discovery of miRNAs as new and important regulators of gene expression is expected to broaden our biological understanding of the regulatory mechanism in muscle by adding another dimension of regulation to the diversity and complexity of ge...
